Effects of Acid-Base on Membrane potential of Rabbit Papillary Muscles

Abstract
The effect of acid-base on the electrical and mechanical properties of myocardial cells were investigated in the isolated right ventricular papillary muscle preparations of rabbits by measuring simultaneously isometric contraction and the action potential by glass microelectrode transducer in acidic or alkali conditions respectively. Results were as follow : 1. The acid condition decreased the isometric contraction and at the sametime increased the time to repolarize 90% of its action potential amplitude.(Repol 90%) from 234¡¾9.1 msec of the control to 244¡¾6.8msec(P<0.01). 2. The alkali condition, on the other hand, increased the isometric contraction and lengthened Repol 90% from 256¡¾49.3 msec of the control to 286¡¾75.2msec(P<0.1) but Repol 95% lengthened more prominently from 281¡¾66.8msec of the control to 329¡¾101.5(P<0.05). The above result suggests that acid and alkali decreased and increased slow inward current respectively to reduce and augment the isometric contraction. In acidic condition, the reduction in delayed outward current might also play a role in augmented action potential duration.
